چکیده
Objective: This study investigated the expression of TRPC1 and TRPC4 in the basilar artery in a rat subarachnoid hemorrhage (SAH) model, and clarified the potential role of TRPC1 and TRPC4 in cerebral vasospasm. Methods: Seventy-two rats were randomized into six groups: control, day 1, day 3, day 5, day 7, and day 14 groups. The day 1, day 3, day 5, day 7, and day 14 groups were the SAH groups. The animals in the SAH groups were injected with autologous blood into the cisterna magna once on day 0, and were sacrificed on days 1, 3, 5, 7, and 14, respectively. The cross-sectional area of the basilar artery was measured, and morphological changes were detected using light microscopy. Immunohistochemisty, PCR, and western blotting were used to assess the expression of TRPC1 and TRPC4. Results: The rat “double-hemorrhage” model of vasospasm was successfully induced. The cross-sectional area of the basilar artery was 57,944±5,581 μm in the control group, and 32,100±2,439 μm, 19,723±2,412 μm, 26,100±2,639 μm, 34,800±2,580 μm, and 57,100±2,579 μm in the day 1, 3, 5, 7, and 14 groups, respectively. The basilar artery exhibited vasospasm after SAH, which became more severe on day 3, and was characterized by arterial narrowing, thickness of arterial wall, and degeneration of the endothelial and smooth muscle cells. In addition, elevated expression of TRPC1 and TRPC4 were detected after SAH, and peaked on day 3. Conclusion: The increased expression of TRPC1 and TRPC4 mirrors the development of cerebral vasospasm in a rat experimental model of SAH. These findings suggest that TRPC1 and TRPC4 may have a role in the pathogenesis of cerebral vasospasm after SAH. [Xiaoou Sun, Shaofeng Yang, Yuan Shi, Chengyuan Ji, Linyuan Wu, Zhong Wang. Potential contribution of SOCC to cerebral vasospasm after experimental subarachnoid hemorrhage in rats.
Cerebral vasospasm (CVS) is the most treatable component of subarachnoid hemorrhage (SAH), which can be reduced by endothelin receptor antagonists. Endothelin-evoked vasospasm is considered to be mediated by Ca(2+) influx in the smooth muscle through voltage-dependent Ca(2+) channel (VDCC) and nonselective cation channels (NSCC). Because VDCC antagonists such as nimodipine have been shown to be...
متن کاملMagnesium sulfate reverses experimental delayed cerebral vasospasm after subarachnoid hemorrhage in rats.
We induced experimental delayed cerebral vasospasm by the intracisternal injection of greater than 0.5 ml blood in 30 rats. Seventy-two hours later the basilar artery was exposed via the transclival approach and photographed at high-power magnification through an operating microscope. We then evaluated the effect of topical (n = 30) and intravenous (n = 20) magnesium sulfate on the spastic arte...
متن کاملChronic cerebral blood flow changes following experimental subarachnoid hemorrhage in rats.
Experimental subarachnoid hemorrhage was induced in 52 adult male Wistar rats by microsurgical transclival basilar artery puncture. Telencephalic blood flow measured in 24 rats with subarachnoid hemorrhage was compared with that in 23 sham-operated rats and 10 unoperated control rats using the [14C]butanol indicator fractionation technique. Telencephalic blood flow was significantly less in the...
متن کاملPotential contribution of matrix metalloproteinase-9 (mmp-9) to cerebral vasospasm after experimental subarachnoid hemorrhage in rats.
This study investigated the possible involvement of matrix metalloproteinase 9 (MMP-9) in cerebral vasospasm (CVS) after subarachnoid hemorrhage (SAH) in rats. The CVS model was established by injection of fresh autologous nonheparinized arterial blood into the cisterna magna. Experiment 1 aimed to investigate the timecourse of the MMP-9 expression in the basilar artery after SAH. In Experiment...
متن کاملCORRELATION BE TWEEN ENDOTHELIAL INJURY AND CEREBRAL VASOSPASM FOLLOWING A DOUBLE SUBARACHNOID HEMORRHAGE IN THE RAT
While a wide array of pathological changes occur in cerebral arteries following subarachnoid hemorrhage (SAH), the most consistent is endothelial damage. Since the endothelium normally modulates reflexes that influence vascular tone, any damage to it may represent a significant contributor to cerebral vasospasm following SAH. This experimental study investigates the correlation between end...
